SIM culture medium is a semi-solid culture medium for microbial identification, and its name comes from the fact that it can carry out three tests simultaneously: sulfide production (S), indole production (I) and mobility (M) detection. The basic components of the culture medium include peptone (providing sulfur-containing amino acids and tryptophan) and a small amount of agar (about 0.4%) to form a semi-solid state. After the puncture inoculation culture, if the bacteria diffuse along the puncture line and make the culture medium turbid, it is judged as dynamic positive; If the sulfur-containing amino acid is decomposed to produce hydrogen sulfide, it will react with iron salt in the culture medium to produce black ferrous sulfide precipitate; After culture, Kovack's reagent is added dropwise, and if a red ring appears on the liquid surface, it is indole positive. The culture medium integrates three key biochemical characteristics, and is especially suitable for rapid identification of Enterobacteriaceae bacteria, with simple operation and high efficiency.
