Lysine iron agar (LIA)
Lysine-iron agar (LIA) is a slant culture medium for identifying intestinal bacteria, especially Salmonella. Its key components include lysine, glucose, ferric ammonium citrate and phenol red indicator. After puncture inoculation and slant culture, it was interpreted by observing the color of slant and the phenomenon of gas production/blackening at the bottom. If bacteria only ferment glucose to produce acid, the slope and upper layer will turn yellow; If there is lysine decarboxylase, lysine will be decomposed to produce alkali, so that the inclined plane will return to red and the upper yellow, which is a typical reaction of Salmonella (K/A, black line). If hydrogen sulfide is produced, it will react with iron salt to form black precipitate. The culture medium can effectively distinguish Salmonella (positive) from Shigella (negative) through the antagonistic reaction between lysine decarboxylation and glucose fermentation.
