XLT4 agar
XLT4 agar is a highly selective separation medium, which is specially used for the separation and identification of Salmonella in food and environmental samples. Its selectivity mainly depends on high concentrations of sodium thiosulfate, ammonium ferric citrate and sodium deoxycholate, which can strongly inhibit the growth of non-target bacteria such as gram-positive bacteria and coliforms. The typical colony of Salmonella on this medium is black or central black, because it produces hydrogen sulfide, which reacts with iron salts to form black precipitate. The fermentation difference between xylose and lactose plays an identification role, and phenol red is used as an indicator. Compared with other media, XLT4 agar can inhibit Proteus and other miscellaneous bacteria more effectively, and also has a good effect on atypical salmonella (such as H2S-producing strains), which has high sensitivity and specificity and is one of the international standard methods.
