NMY51-MEL1 Chemically Competent Cell
NMY51-MEL1 Chemically Competent Cell
The DUAL membrane system is a specialized detection technology for screening interactions between transmembrane proteins, utilizing a yeast two-hybrid system with a separated ubiquitin system to directly detect interactions between membrane proteins under natural conditions. This system employs the NMY51 yeast strain, which can be directly transformed with plasmids for protein interaction validation or library screening. Based on the Punter Bio NMY51 strain, the MEL1 reporter gene is introduced, with the transformation marker being trp1, leu2-3, and the reporter genes being HIS3, ADE2, lacZ, and MEL1. The first step involves selective growth screening using the nutritional-deficient reporter genes (HIS3, ADE2), followed by quantitative or semi-quantitative screening via β-galactose analysis using the lacZ reporter gene (or quantitative or semi-quantitative screening via α-galactose analysis using the MEL1 reporter gene). The four independent reporter genes are regulated by different promoters to reduce the likelihood of false positives. This system can utilize four bait plasmids: pBT3-N, pBT3-C, pBT3-SUC, and pBT3-STE, with all screening markers being LEU; and four prey plasmids: pPR3-C, pPR3-N, pPR3-SUC, and pPR3-STE, with all screening markers being TRP. The NMY51-MEL1 competent cells are produced through a specialized process and can be stored at -80°C for 12 months. The detection transformation efficiency of the pGADT7 plasmid is>104 cfu/μg DNA.
