I-top medium

J-top culture medium is a kind of soft agar covering layer specially used for bacterial conjugation test. Its components are liquid or semi-solid (low-concentration agar) culture medium with rich nutrients (such as peptone and yeast extract), which is used to mix and cultivate donor bacteria and recipient bacteria. In the conjugation experiment, donor and recipient bacteria with different genetic markers (such as antibiotic resistance) were mixed in a small amount of this medium, and then poured on the V-B agar plate with the lowest nutrition. The soft texture and rich nutrition of H- top culture medium provide an ideal environment for bacteria to contact, pair and transfer plasmid DNA. After culture, only the recipient bacteria (that is, zygotes) that have successfully joined and obtained the plasmid of the donor bacteria can grow colonies on the bottom layer or selection plate containing specific antibiotics, thus proving the occurrence of joining.
