7.5% sodium chloride broth
7.5% sodium chloride broth is a highly selective liquid medium for screening and enriching salt-tolerant microorganisms, especially for the preliminary screening of Staphylococcus. Its core is high concentration of sodium chloride (7.5%), which can effectively inhibit the growth of most non-salt-tolerant bacteria (such as Escherichia coli and Streptococcus), while salt-tolerant bacteria such as Staphylococcus and Micrococcus can grow and reproduce normally under this condition. Peptone and beef extract in the culture medium provide basic nutrition for the target bacteria. After inoculation, the sample is cultured at 35-37℃ for 18-24 hours. If the culture medium is turbid, it indicates that there may be salt-tolerant bacteria such as Staphylococcus, which need to be further streaked and inoculated on selective solid culture medium (such as mannitol salt agar) for confirmation. This culture medium is an important screening tool for food hygiene inspection and detection of pathogenic staphylococci in clinical samples.
