The preparation method of Norstog medium, a classic medium specifically designed for anther culture of gramineous crops (such as barley, wheat. It was developed by plant physiologist K. Norstog and is very effective in promoting the embryogenesis of microspores. Norstog medium usually uses 612% sucrose, and the higher osmotic pressure helps microspores to switch from the gametophytic developmental pathway to the sporophytic pathway (i.., to produce embryoids). Compared with MS medium, it has a lower concentration of inorganic salts, which is more suitable for anther culture, and Fe-EDTA (chelated iron) is used to ensure the effectiveness of iron.
