Mannitol in the culture medium is used as fermentable carbon source and phenol red is used as pH indicator. The colony of Bacillus cereus which does not ferment mannitol is gray or colorless, and the color of the surrounding culture medium remains unchanged (pink/alkaline); However, the mixed bacteria that ferment mannitol produce acid and turn yellow The key distinguishing feature is yolk reaction: Bacillus cereus produces lecithin enzyme, which decomposes yolk to form opaque white precipitation ring around colony. This characteristic is an important index to identify the strain.
