Iger LT100 Broth is a special liquid culture medium for selective enrichment of Legionella in water. Its ingredients are optimized, including yeast extract, ACES buffer, α-ketoglutaric acid, etc., which provide special nutrients (such as L- cysteine and iron ions) needed for the growth of Legionella and maintain a stable pH environment (about 6.9). The key selectivity lies in the combination of glycine, polymyxin B, vancomycin and cycloheximide (CYE), which can effectively inhibit the growth of most miscellaneous bacteria (including Gram-positive bacteria, Gram-negative bacteria and fungi). Samples (such as water concentrated solution) are inoculated and cultured at 35-36℃, which usually takes 2-5 days. This culture medium provides a high-purity enrichment foundation for subsequent plate separation and culture, and significantly improves the detection sensitivity, which is the key enrichment step in the standard method of Legionella monitoring in water environment.
