3% sodium chloride trisaccharide iron agar
3% sodium chloride trisaccharide iron agar
3% sodium chloride trisaccharide iron agar is a selective identification medium specially used to identify Vibrio, especially Vibrio parahaemolyticus. Its basic composition is similar to that of common iron trisaccharide agar, which contains peptone, three sugars (lactose, sucrose and glucose) and phenol red indicator. The key difference is that 3% high concentration sodium chloride is added to create a hypertonic environment and inhibit the growth of non-halophilic bacteria, so as to preliminarily screen halophilic Vibrio. After inoculation, the results were judged by observing the color change of inclined plane and bottom layer and the gas production. If bacteria can ferment glucose to produce acid, it will make the bottom layer yellow; If lactose and/or sucrose are fermented at the same time, the inclined plane will also turn yellow; If it can't grow because of high salt, the culture medium remains red. Vibrio parahaemolyticus is characterized by slant red (lactose and sucrose are not fermented), yellow bottom layer (glucose is fermented) and no gas production, and at the same time, the bottom of the culture medium turns black due to hydrogen sulfide production. The culture medium integrates high salt selectivity, sugar fermentation, hydrogen sulfide production and other biochemical reactions, and is an important tool in aquatic product inspection and intestinal pathogen screening.
