42℃ growth medium
The growth medium at 42℃ is not a medium with fixed components, but an identification method with high temperature as the key selection condition. Its foundation can be general liquid culture medium such as common nutrient broth, tryptone soybean broth, etc. The core principle of this method is that the bacteria to be tested are inoculated into the culture medium and then cultured in a high temperature environment of 42℃. This temperature is a severe survival challenge for most mesophilic bacteria (the optimum growth temperature is about 37℃), which will strongly inhibit their growth and even lead to death. Therefore, 42℃ culture is mainly used in two situations: one is to identify fecal coliforms as a confirmation test of Escherichia coli, because Escherichia coli can ferment lactose to produce gas at 42℃, while other enterobacteria can't; Second, the optimal growth temperature for screening thermophilic bacteria or some specific pathogenic bacteria, such as Campylobacter jejuni, is 42-43℃. By observing whether it grows at 42℃, the identification range can be effectively narrowed, which is a simple and important selective identification method.
