AMCS is widely present in the mitochondrial matrix of animals, plants, microorganisms, and cultured cells, and participates in the regulation of the tricarboxylic acid cycle together with citrate synthase (CS). MCS catalyzes propionyl-CoA and oxaloacetate to produce methyl.citryl-CoA, which is further hydrolyzed to produce methylcitric acid; this reaction promotes the conversion of colorless DTNB into yellow TNB, which has a characteristic absorbance at 412 nm.
