Service Introduction

The yeast sugar transporter detection experiment refers to using yeast sugar-deficient mutants to test whether a protein expressed in yeast can transport the relevant sugars into yeast cells for utilization.

Yeast Hexose Transport Activity Detection

Specifically designed for studying yeast hexose (such as glucose and fructose) transporter protein functions, it is based on the principles of heterologous complementation and growth phenotype verification in yeast. This includes a core hexose transport-deficient Saccharomyces cerevisiae strain (such as EBY.VW4000), the pDR196 high-efficiency expression vector, and supporting culture medium reagents. The deficient strain, lacking all endogenous hexose transport proteins, cannot absorb hexose by itself and can only grow on maltose-containing media. After introducing the target transporter gene into the strain, an active protein can mediate hexose uptake, allowing the strain to grow normally on a medium with hexose as the sole carbon source. This specifically excludes interference from non-hexoses and is suitable for verifying hexose transporter protein functions, screening metabolic genes, and studying transport mechanisms.

Yeast Sucrose Transport Activity Detection

Specifically designed for studying yeast sucrose metabolism and transporter protein functions, it is based on the principle of heterologous expression in yeast. This includes a sucrose transport-deficient yeast strain (SUSY7/ura3), expression vectors, and supporting selective media reagents. The yeast mutant (SUSY7/ura3) cannot secrete invertase but can express a gene encoding a plant-derived cytosolic sucrose synthase, enabling yeast to metabolize sucrose intracellularly. This mutant strain, together with cDNA plasmids constructed on the yeast expression vector, is transformed, and the transformants are selected based on their growth ability on media with sucrose as the sole carbon source. This method intuitively reflects sucrose transport capability, excluding interference from other sugars, and is suitable for transporter protein function verification, sucrose metabolism gene screening, and regulatory mechanism studies.

Service Advantages

▶Standardization of mutant strains: Use internationally recognized strains to ensure experimental reproducibility, meeting the requirements of journals such as Nature and Plant Cell.

▶Substrate specificity verification: Use monosaccharide/disaccharide gradient media to rule out cross-reactions.

▶Data traceability: Provide original growth images, quantitative curves, and statistical analysis reports.

▶Custom service capabilities: Detection conditions can be optimized according to customer needs to suit the functional identification of special proteins.
