Agar No.4 is a strong selective solid medium, which is specially used for the separation and culture of Shigella and Salmonella. Its selectivity mainly depends on three inhibitors: bile salt, sodium citrate and brilliant green. They can strongly inhibit the growth of Gram-positive bacteria, Escherichia coli and other miscellaneous bacteria, while allowing the growth of Shigella and Salmonella. Lactose in the culture medium and indicator neutral red are used for preliminary identification: the colonies of bacteria that can ferment lactose (such as Escherichia coli if not completely inhibited) are red, while Shigella and Salmonella that do not ferment lactose form transparent or translucent colorless colonies. Therefore, on this plate, colorless and transparent colonies are suspected pathogens. Agar 4 has high selectivity and intuitive identification effect, and is one of the important media commonly used in the detection of intestinal pathogens, especially in the diagnosis of bacillary dysentery.
