pBAD33 vector

The pBAD33 utilizes a very stringent "araBAD promoter/AraC regulatory system". In the absence of L-arabose, the AraC protein represses the promoter, resulting in very low basal leaky expression; upon addition of L-arabinose, it binds to AraC changes its conformation, thereby initiating the transcription of downstream genes. This "switch" property is particularly advantageous for the expression of proteins that are toxic to the cell or could potentially normal growth. The expression level can be precisely controlled by adjusting the concentration of L-arabinose, allowing for expression from low levels to high amounts. It is used for protein in E. coli, including functional studies, antibody production, and protein purification. Additionally, it is also commonly used in gene complementation experiments.
