pK18-Mob SacB is a derivative of the r plasmid and has a suicide nature, meaning that it cannot replicate autonomously in certain specific bacteria. This is because the replication proteins required for initiating the plasmid's replication are usually absent in these bacteria. Once pK18-Mob SacB enters the host cell, it either cannot replicate and is eliminated, or, under external selective pressure, integrates into the host chromosome and replicates along with it.
