Sodium chloride sucrose agar
Sodium chloride sucrose agar is a selective medium for the identification of pathogenic Vibrio, especially for the preliminary identification of Vibrio parahaemolyticus. Its formula contains peptone, high sodium chloride concentration (usually 3%) to inhibit non-halophilic bacteria, and sucrose and phenol red indicators. Most Vibrio can ferment sucrose to produce acid, which turns the culture medium around the colony yellow. The key feature of pathogenic Vibrio parahaemolyticus is that it does not ferment sucrose, so it forms a green or blue colony (alkaline reaction) that does not change color, which is in sharp contrast with the surrounding miscellaneous bacteria that may turn yellow. This sucrose negative reaction is an important identification basis, which makes the culture medium have practical value in seafood safety detection and clinical diagnosis.
