PALCAM agar
PALCAM agar is a highly selective chromogenic medium for the separation and identification of Listeria. Its name comes from the main selective components: polymyxin B, lithium chloride, ceftazidime and acridine yellow. These inhibitors can effectively inhibit the growth of most gram-positive bacteria and gram-negative miscellaneous bacteria. The identification system of the culture medium is based on two biochemical reactions: Listeria can ferment aescin, which leads to the blackening around its colony (forming a black halo); And can hydrolyze mannitol, so that the colonies are grayish green because of the surrounding acid production and phenol red indicator. Therefore, on PALCAM agar, the typical listeria is grayish green with a black halo in the center. The culture medium has high selectivity and intuitive identification characteristics, and is one of the standard tools for the detection of Listeria in food and environment.
