PALCAM broth

PALCAM broth is a liquid medium for selective enrichment of Listeria monocytogenes. Its composition corresponds to the selective system of PALCAM agar, and it is often used in the pre-separation step after enrichment. Broth contains the same selective inhibitors as agar, including polymyxin B, lithium chloride, ceftazidime and acridine yellow, which can effectively inhibit the background of miscellaneous bacteria. At the same time, it also contains aescin and mannitol as indicator system. The fermentation of aescin by Listeria will make the broth black (forming aescin-iron complex). This characteristic can provide a preliminary positive indication in the enrichment stage. Usually combined with PALCAM agar, the enriched broth is streaked and inoculated on the plate for final separation and confirmation, thus improving the efficiency of detecting Listeria from complex samples.
