Half-Fraser medium
Half-Fraser medium, that is, semi-Frazier medium, is the first step of the two-step enrichment method for detecting Listeria. Its formula is half of the concentration of basic Fraser medium, and contains nutrients such as peptone and yeast extract, as well as selective inhibitors (such as nalidixic acid and acridine yellow). This "half-strength" design aims to provide a mild and selective environment, which can not only initially inhibit some miscellaneous bacteria, but also avoid excessive inhibition of Listeria which may be damaged, especially conducive to the recovery and initial proliferation of damaged cells. After inoculation, the samples were cultured at 30℃ for 24 hours to complete the initial enrichment, and then transferred to Fraser broth with more selectivity for secondary enrichment. This two-step method significantly improves the detection rate of Listeria, which is the key link of the international standard method.
