Oxford agar (OXA) is a highly selective medium, which is specially used to isolate and identify Listeria monocytogenes. Its selectivity mainly depends on the combination of antibiotics (such as colistin, polymyxin B, cefthiophene, etc.), which can effectively inhibit most gram-positive bacteria and gram-negative miscellaneous bacteria. The culture medium contains aescin and iron salt. Listeria can hydrolyze aescin to produce black products, which makes black halo around the colony, which is the key identification feature. In addition, Listeria can also ferment mannitol in the culture medium, resulting in a unique black or brown-green colony due to acid production around it. OXA is highly selective, and it is one of the standard media for isolating Listeria in food microbiological examination.
