Semi-solid dynamic medium is a classic identification medium for observing bacterial motility. Its composition is similar to that of ordinary broth, but a low concentration of agar (usually 0.3%-0.4%) is added to form a soft gelatinous semi-solid state. During inoculation, a puncture needle is used to vertically pierce the center of the culture medium. After culture, unpowered bacteria only grow along the puncture line with clear lines; Dynamic bacteria can swim around the puncture line by flagella, making the whole culture medium cloudy. This method is a simple, intuitive and reliable experiment to judge whether bacteria have flagella and can exercise, and it is of great value in the preliminary identification of bacteria such as Enterobacteriaceae.
