pHiS2 Vector
pHiS2 vector serves as both a “reporter vector” and a “bait vector” in the yeast one-hybrid experiment. The core involves the use of “HIS3” as a reporter gene. The promoter to be studied (i.e., a DNA sequence that may be bound by a protein) cloned into the multi-cloning site of the vector. This multi-cloning site is positioned upstream of the minimal promoter of the HIS3 reporter gene. If a protein in yeast cells specifically binds to the DNA sequence you have inserted, this binding will activate the transcription of the HIS3 reporter gene.
