Sorbitol MacKay agar base (SMAC)
Sorbitol MacKay agar base (SMAC)
Sorbitol MacKay agar base (SMAC) is a selective identification medium, which is specially used to isolate and preliminarily identify pathogenic Escherichia coli, especially serotype O157:H7. Based on MacKay agar, which contains bile salts and crystal violet, it can inhibit the growth of Gram-positive bacteria, thus selectively promoting the growth of Gram-negative Enterobacter. The key identification component is sorbitol. The vast majority of Escherichia coli can rapidly ferment sorbitol to produce acid, making the colony pink; However, pathogenic strains such as O157:H7 lack this ability and do not ferment sorbitol, thus forming colorless or neutral colonies. This remarkable color difference makes SMAC an important tool for screening pathogenic bacteria related to hemorrhagic enteritis in clinical microbiology and food safety detection.
