β-GAL is widely present in animals, plants, microorganisms, and cultured cells, and it can catalyze the hydrolysis of the β-galactosidic bond in β-galactoside compounds. In addition, it also has trans-galactosylation activity. β-GAL not only releases stored energy for the rapid growth of plants but also catalyzes the hydrolysis of terminal galactose residues in polysaccharides, glycoproteins, and galactolipids during normal polysaccharide metabolism, cell wall component metabolism, and cell wall degradation during senescence, releasing free galactose. β-GAL breaks down p-nitrophenyl-β-D-galactopyranoside to produce p-nitrophenol, which has a maximum absorption peak at 420nm. The β-GAL activity can be calculated by measuring the rate of increase in absorbance.
