Yeast Transcription Activity Detection Kit
Yeast Transcription Activity Detection Kit
Herpes simplex virus protein 16 (VP16) is a transcriptional activator that can activate the expression of downstream genes. By fusing the VP16 transcriptional activation domain with GAL4BD and expressing it, the pGBKT7-VP16 (BD-VP16) vector was constructed and transformed into Y2HGold. The transcriptional activation function of VP16 will activate the expression of yeast reporter genes HIS3 and ADE2, allowing normal growth on histidine- and adenine-deficient media. Transcription factor X is fused separately with BD and BD-VP16 for expression. If transcription factor X has a transcriptional activation effect on downstream genes, it will activate their expression. Compared with the control strain containing BD, the strain containing BD-X can grow on deficient media. If transcription factor X has a transcriptional repression effect on downstream genes, it will partially inhibit the activation of downstream genes by VP16. Compared with the control strain containing BD-VP16, the strain containing BD-VP16-X will inhibit the activation of reporter genes downstream, thereby suppressing growth on deficient media.
