AH109-GAL4 Yeast Two-Hybrid Library Screening kit (Plasmid Transformation Method)
AH109-GAL4 Yeast Two-Hybrid Library Screening kit (Plasmid Transformation Method)
![]() Many eukaryotic site-specific transcriptional activators typically have two separable domains: the DNA-binding domain (BD) and the transcriptional activation domain (AD). These two domains each have distinct functions and do not interfere with each other, but a functional activator that can activate the expression of a specific gene must contain both domains; otherwise, it cannot perform its activation role. When the BD of an activator from one source binds with an AD, it specifically activates the gene whose expression is recognized by the BD. Based on this principle, two proteins to be tested can be fused separately with these two domains and co-expressed in the same yeast cell. If the two proteins interact with each other, they can bridge AD and BD together to form a complete transcriptional activator and thus activate the corresponding reporter gene expression. By measuring the phenotype of the reporter gene, it is easy to determine whether a molecular interaction occurs between the proteins being tested.
