The Y187-pHis2 yeast one-hybrid system requires the use of two plasmids, pHis2 and pGADT7-Rec2, in combination. The selection marker for the pHis2 plasmid is Trp, which is used to express pBait-His2 (the full- promoter is cloned into pHis2); the selection marker for the pGADT7-Rec2 plasmid is LEU, which is used to express the fusion protein AD (amino acids 768~881 at the C-terminus of GAL4) and the target protein (Prey). Principle of Y187-pHis2 yeast one-hybrid system: When the target protein (Prey) binds to the bait sequence (BaitDNA), GAL AD will activate the expression of HIS, which is able to grow in the medium containing 3-AT, thereby determining that there is an interaction between DNA and protein.
