Y187-pHis2 Yeast One-Hybrid Library Screening kit

The Y187-pHis2 yeast one-hybrid system requires the use of two plasmids, pHis2 and pGADT-Rec2, in combination. The selection marker for the pHis2 plasmid is Trp, which is used to express pBait-His2 (the full- promoter is cloned into pHis2); the selection marker for the pGADT7-Rec2 plasmid is LEU, which is used to express the fusion protein AD (amino acids 768~881 at the C-terminus of GAL4) and prey protein (Prey). Principle of the187-pHis2 yeast one-hybrid system: When the prey protein (Prey) binds to the bait sequence (BaitDNA), GAL4 will activate the expression of HIS, which is able to grow on the medium containing 3-AT, thereby determining that there is an interaction between DNA and protein.
