Membrane Yeast Two-Hybrid Screening Kit Plus
Membrane Yeast Two-Hybrid Screening Kit Plus
![]() The membrane yeast two-hybrid system: Based on the split-ubiquitin-mediated membrane protein yeast two-hybrid system, which can be used to detect interactions between membrane protein-membrane protein or membrane protein-soluble protein, and can also be applied for library screening. The ubiquitin molecule is divided into two structural parts, Cub and NubG, each fused to a known bait protein and a prey protein, respectively, and properly expressed in the membrane. The Cub end is linked to the transcriptional activator LexA-VP16 on the cytoplasmic side. When the known bait protein and prey protein interact, the interaction brings NubG and Cub close together spatially, forming a complete ubiquitin molecule. This leads to the cleavage of the cytoplasmic transcriptional activator LexA-VP16 by UBPs, releasing LexA-VP16 to enter the nucleus and activate the expression of His3, Ade2, and LacZ. Proteins that interact can be screened using appropriate selective media and β-galactosidase colorimetric assay kits.
