NMY51 DUALhunter System Yeast Two-Hybrid interaction proving Kit
NMY51 DUALhunter System Yeast Two-Hybrid interaction proving Kit
![]() The DUALhunter system is based on the DUALmembrane system, with appropriate modifications, i.e., by fixing cytoplas proteins (or nuclear proteins) to the cell membrane through a small membrane protein (yeast ER protein Ost4), which facilitates detection (see figure). First, the-terminus of the bait protein (Bait-Cub-LexA-VP16) is fused with a small membrane anchor protein (Ost4) to a fusion protein (Ost4-Bait-Cub-LexA-VP16); next, the target protein is fused to the N-terminus of ubiquitin NubG portion; if bait and prey interact, the two parts of ubiquitin, NubG and Cub, are brought close together and separated ubiquit is reconstituted, recognized by UBPs, leading to the dissociation of LexA-VP16, which enters the nucleus and thus activates the transcription of the reporter.
