Y2HGold-GAL4 Yeast Two-Hybrid interaction proving kit
Y2HGold-GAL4 Yeast Two-Hybrid interaction proving kit
![]() Basic Principle of Yeast Two-Hybrid: Many site-specific transcriptional activators in eukaryotes typically have two separable structural domains: a DNA-binding domain (BD) and a transcriptional activation domain (AD). Each of these domains has its own function and does not interfere with the other. However, a complete activator that can specifically activate gene expression must contain both domains simultaneously; otherwise, it cannot perform its activating function. When the BD of an activator from one source combines with the AD from another, it specifically activates the gene expression bound by BD. Based on this principle, two proteins of interest can be fused separately to these two domains and co-expressed in the same yeast cell. If the two proteins interact, they will act as a bridge to bring AD and BD together, forming a complete transcriptional activator and activating the corresponding reporter gene expression. By measuring the reporter gene phenotype, it is easy to determine whether there is an interaction between the proteins of interest.
