Y2HGold-GAL4 System Yeast Two-Hybrid Screening Library Kit (Plasmid Transformation Method)
Y2HGold-GAL4 System Yeast Two-Hybrid Screening Library Kit (Plasmid Transformation Method)
Principle of the yeast two-hybrid system: Many site-specific transcription activators in eukaryotes generally have two separable domains: the DNA-binding domain (BD) and the transcriptional activation domain (AD). These two domains function independently without affecting each other, but a complete activator that triggers specific gene expression must contain both domains; otherwise, it cannot perform its activation function. When the BD of one activator binds to the AD from another source, it specifically activates the expression of genes to which the BD binds. Based on this principle, two proteins to be tested can be fused separately with these two domains and co-expressed in the same yeast cell. If the two proteins interact with each other, they will bring AD and BD together through the protein bridge, forming a complete transcriptional activator and activating the corresponding reporter gene expression. By measuring the reporter gene phenotype, it is easy to determine whether the proteins under investigation interact at the molecular level.
