Nitrate broth
Nitrate broth is a biochemical test medium for detecting nitrate reduction ability of microorganisms. Its main components include peptone, beef extract powder to provide basic nutrition, and key reagent potassium nitrate. After inoculating bacteria and culturing, sulfanilamide and α-naphthylamine reagents are added for detection. If the culture solution is red (diazo reaction), it indicates that bacteria reduce nitrate to nitrite, which is a positive reaction. If there is no discoloration, it may be negative or nitrate is further reduced, which needs to be verified by adding zinc powder. This test is an important basis for distinguishing Enterobacteriaceae and other bacteria
