EMMS Medium
![]() EMM medium (Edinburgh Minimal Medium), also known as Edinburgh Minimal Medium, is a synthetic defined medium used for the nutritional growth of Schizosaccharomyces pombe, providing glucose, minerals, vitamins, and trace elements. For culturing to inhibit the NMT promoter, 15 μM thiamine can be added to EMM medium, and for moderate inhibition, 0.05 μM thiamine is added. Adenine content is reduced to 10 mg/L to observe the red phenotype in ade6 mutants. Low-glucose EMM medium is used for protoplast preparation and yeast transformation. EMMG medium is used for liquid culture of Schizosaccharomyces pombe diploid spores. This medium is based on EMM medium, with L-glutamic acid replacing ammonium chloride as the nitrogen source. EMMS medium has additional sorbitol added and is used to study the nmt1 promoter in transformed fission yeast.
