pSilencer-U5-puro is an RNA interference (RNAi) dedicated vector developed by Ambion (now part of Thermo Fisher). Its core function is to stably express small interfering RNA (siRNA) or short hairpin RNA (shRNA) in mammalian cells, specifically silencing target genes through RNA interference. It also includes a puromycin selection marker, allowing efficient selection of stably silenced cell lines. Its design feature is the use of the U6 promoter to drive small RNA transcription, making it suitable for long-term gene silencing studies.
