pCAMBIA1307-6×myc-YNE plant dual-expression vector
pCAMBIA1307-6×myc-YNE plant dual-expression vector
pCAMBIA1307-6×myc-YNE is a plant expression vector constructed based on the pCAMBIA1307 backbone. It contains an E. coli origin of replication (such as ColE1 ori), allowing high-copy replication in E. coli, which facilitates vector amplification and manipulation. At the same time, it also has an Agrobacterium origin of replication (such as oriV), enabling the vector to stably exist and replicate in Agrobacterium, thereby supporting Agrobacterium-mediated plant transformation. Additionally, the vector usually carries prokaryotic selection marker genes, such as the kanamycin resistance gene (nptII), which is used to select E. coli and Agrobacterium containing the vector. The plant selection marker gene is generally the hygromycin resistance gene (hpt), which can be used to select plant cells or plants that have successfully incorporated the vector. A key feature of the vector is the introduction of a 6×myc tag sequence in the multiple cloning site region. This tag consists of six tandem myc tags (amino acid sequence EQKLISEEDL), which can specifically bind to anti-myc antibodies, facilitating the detection and analysis of the fusion protein. Downstream of this tag is the YNE target gene sequence.
