Phospholipase A2 (EC3.1.1.4) is a lipid acyl hydrolase at the sn-2 position of phospholipids, which is widely present in animal and plant tissues, bacteria, and nuclear secretions, and participates in physiological processes such as fat digestion, sperm maturation, cell signal transduction, lipid peroxidation repair, and host response.

Phospholipase A2 acts on 2-thiocecedecyl ethylphosphocholine (HEPC) to produce a free sulfhydryl group, which reacts with DTNB to form a yellow substance with a characteristic absorption peak at 412 nm.
