Columbia blood agar base is a universal basic medium with extremely rich nutrition, which is mainly used for the separation and culture of fastidious bacteria, especially after adding 5%-10% defibrinated sheep blood to make Columbia blood agar, which can support the growth of most pathogenic bacteria (including streptococcus, pneumococcus, etc.). Its basic components include casein trypsin digest, heart and brain hydrolysate, corn starch, sodium chloride, etc., which can provide sufficient nitrogen source, carbon source, vitamins and minerals. The added blood not only provides additional growth factors (such as X factor and V factor), but also can directly observe the hemolytic characteristics of colonies (α, β, γ hemolysis), which is an indispensable basic culture medium in clinical microbiology.
