DNA enzyme methyl green agar base is a differential medium for detecting the activity of microbial DNA enzyme. Its basic components include casein hydrolysate, plant peptone and other nitrogen and carbon sources, and its key components are high molecular weight DNA as reaction substrate and methyl green as indicator. Bacteria with dnase activity (such as Staphylococcus aureus, Serratia, etc.) can cut the long DNA chain, resulting in the dissociation and fading of methyl green indicator bound to DNA, forming a clear colorless transparent ring around the colony. This test is an important index to identify Staphylococcus aureus and some Gram-negative bacilli, which is widely used in clinical and food microbiology identification.
