Glutathione S-transferases (GSTs) are a family of proteins with multiple physiological functions, primarily present in the cytoplasm. As a crucial component of the body's detoxifying enzyme system, GSTs mainly catalyze the covalent binding between various chemical substances (and their metabolites) and the sulfhydryl group of glutathione (GSH). This reaction converts electrophilic compounds into hydrophilic substances, which can be easily excreted through bile or urine, thereby achieving the goal of degrading and eliminating various potential or toxic substances from the body. Therefore, GSTs play a vital biological role in protecting cells from damage caused by electrophilic compounds. Additionally, due to their glutathione peroxidase (GSH-Px) activity, GSTs are also referred to as non-selenium-dependent glutathione peroxidases (non-Se GSH-Px) and possess the function of repairing macromolecules (such as DNA and proteins) damaged by oxidation. It should be noted that the reactions catalyzed by GSTs reduce the content of GSH but do not increase the content of oxidized glutathione (GSSG).

GSTs catalyze the binding of GSH to 1-chloro-2,4-dinitrobenzene (CDNB), and the reaction product has a light absorption peak at a wavelength of 340 nm. The activity of GSTs can be calculated by measuring the rate of increase in absorbance at the wavelength of 340 nm.
