Glutathione Reductase (GR) is a flavoprotein oxidoreductase widely present in eukaryotes and prokaryotes, and it is one of the key enzymes in the glutathione redox cycle (typically, GR is replaced by Thioredoxin Reductase (TrxR) in insects). GR catalyzes the reduction of oxidized glutathione (GSSG) to reduced glutathione (GSH) using nicotinamide adenine dinucleotide phosphate (NADPH) as a cofactor, which helps maintain the intracellular GSH/GSSG ratio. GR plays a crucial role in reactive oxygen species (ROS) scavenging during oxidative stress responses; in addition, it is also involved in the ascorbate-glutathione cycle pathway.

GR can catalyze the reduction of GSSG by NADPH to regenerate GSH, and at the same time, NADPH is dehydrogenated to form nicotinamide adenine dinucleotide phosphate (NADP+). NADPH has a characteristic absorption peak at 340 nm, whereas NADP+ shows no absorption at this wavelength. The activity of GR can be calculated by measuring the dehydrogenation rate of NADPH, which is determined by monitoring the rate of decrease in absorbance at 340 nm.
