γ-glutamylcysteine Ligase (GCL) Activity Assay Kit

Glutamate-Cysteine Ligase (GCL) is the rate-limiting enzyme in glutathione (GSH) synthesis, and GSH exerts a feedback inhibitory effect on GCL. The expression of the GCL gene is regulated by a variety of factors, such as oxidants, antioxidants, growth factors, and inflammatory factors. The level of GCL activity has a significant impact on the GSH content and the GSH/oxidized glutathione (GSSG) ratio. In the presence of adenosine triphosphate (ATP) and magnesium ions (Mg2⁺), GCL catalyzes the synthesis of γ-glutamylcysteine from glutamate and cysteine; meanwhile, ATP undergoes dephosphorylation to produce inorganic phosphate molecules. The activity of GCL can be calculated by measuring the rate of increase in inorganic phosphate.
