pLKO-Tet-on is a tool vector that combines a tetracycline-inducible system with a lentiviral vector. Its core function is to regulate the expression of shRNA through tetracycline (or its analog doxycycline, Dox), achieving spatiotemporal-specific silencing of target genes. It is suitable for conditional gene knockdown studies in mammalian cells (especially hard-to-transfect cells) and offers unique advantages in dynamic analysis of gene function, studies of toxic genes, and construction of disease models.
