pTCK303 Vector

The pTCK303 vector is introduced into monocot plant cells via Agrobacterium-mediated transformation. After Agrobacterium infects the plant cells, the T-DNA (transferable DNA) region on the vector is transferred and integrated into the plant genome. Subsequently, the hairpin structure expression cassette is transcribed into RNA molecules with a hairpin structure. These RNA molecules are recognized and cleaved by the cell's RNA interference machinery, producing small interfering RNAs (siRNAs). The siRNAs associate with relevant protein complexes to form the RNA-induced silencing complex (RISC). RISC can recognize and bind to mRNA sequences complementary to the hairpin structure, leading to their degradation and achieving the silencing effect on the target gene.
