Pepsin is secreted by the chief cells of the gastric mucosa and breaks down proteins in food into small peptide fragments. It is generally used for the differentiation of neurogenic hypoacidity. A decrease in pepsin secretion can also be caused by conditions such as chronic gastritis, chronic gastric dilatation, and chronic duodenitis.

Pepsin can catalyze the hydrolysis of hemoglobin, and the product tyrosine has a characteristic absorption peak at 275 nm. The enzyme activity is calculated by measuring the change in absorbance.
