Sulfite-polymyxin-sulfadiazine agar base (SPS) is a solid medium for selective isolation and enumeration of Clostridium perfringens. Its basic ingredients include peptone and yeast extract to provide nutrition, and the key selective system is composed of sodium sulfite, polymyxin B and sulfadiazine, which can effectively inhibit the growth of most gram-negative bacteria, gram-positive bacteria and Proteus. Under anaerobic conditions, the reduced sulfite of Clostridium perfringens reacts with ammonium ferric citrate in the culture medium to form insoluble black iron sulfide precipitate, which leads to black colonies. This culture medium is widely used for the detection of Clostridium perfringens in food, water quality and environmental samples.
