Iron sulfite agar

Iron sulfite agar is a solid medium for selective separation and enumeration of sulfite-reducing Clostridium, especially Clostridium nigricans. Its basic components include peptone and yeast extract to provide basic nutrition, while the key components are sodium sulfite and ammonium ferric citrate. Under anaerobic conditions, Clostridium sulfite can reduce sulfite to sulfide, which reacts with iron ions to form insoluble black iron sulfide precipitate, resulting in black colonies. Using this characteristic reaction and the coagulation of agar, the culture medium realizes the separation, preliminary identification and counting of target bacteria, and is widely used in the hygienic microbiological examination of water quality (especially drinking water) and food.
