The soluble protein content of samples is often used for enzyme activity calculation. Additionally, soluble protein content is also applied in the quality analysis of food and other products. Under alkaline conditions, cysteine, cystine, tryptophan, tyrosine, and peptide bonds in proteins can reduce Cu2⁺ to Cu⁺; two molecules of BCA (Bicinchoninic Acid) bind to Cu⁺ to form a purple complex, which has an absorption peak in the range of 540–595 nm, with the strongest absorption peak at 562 nm.
