The soluble protein content of a sample is often used for enzyme activity calculation. Additionally, soluble protein content is also applied in quality analysis of food and other products. In a strongly alkaline solution, biuret reacts with CuSO4 (copper sulfate) to form a purple complex; the intensity of the purple complex is proportional to the protein concentration and is independent of the protein’s molecular weight and amino acid composition. Therefore, this method can be used to determine protein content. The detection range of this method is 1–10 mg of protein, making it suitable for samples with high protein concentration, especially animal-derived materials.
